چکیده

Crystalline NiS, CuS, CoS2, and CdS particles were actively phagocytosed by cells and potently induced morphological transformation of Syrian hamster embryo cells in a concentra tion-dependent fashion. In contrast, the respective amorphous metal sulfide particles (amorphous NiS, CuS, CoS, and CdS) were not as actively phagocytosed by cultured cells and, in comparison to the crystalline form of these compounds, in duced considerably less morphological transformation at both cytotoxic and noncytotoxic exposure levels. Chemical reduc tion of positively charged amorphous NiS with LiAIH4 resulted in active phagocytosis of these particles which was also asso ciated with enhancement of cellular transformation. Crystalline but not amorphous NiS caused considerable strand breaks in the DNA of Chinese hamster ovary cells following 2 to 3 hr exposure at 10 jug/ml as determined by alkaline sucrose gra dient techniques with subsequent determination of DNA molec ular weight. Phagocytized inert particles such as latex beads did not induce transformation or DNA damage, suggesting that genotoxic dissolution products such as Ni2+ rather than the phagocytized particles are responsible for the observed DNA damage and cellular transformation. NiCI2 was about one-third to one-half as potent in inducing cellular transformation com pared to crystalline NiS on a weight basis. These results correlate the selective phagocytosis of crystalline metal sulfides to their more potent activity in the induction of cellular transformation.

Modeling the distribution of deposited energy by alpha particles from Radon 223 decay and its effect on DNA

The ionizing radiations, through physical and chemical processes, lead to simple and complex single- and double- strand breaks, as well as base lesions to the DNA. In this study, taking into account all the physical and chemical processes involved in the interaction of ionizing radiation with matter, the initial damage induced to DNA was evaluated for 5.7 MeV alpha-rays from Radon 223 isotope....

Simulation of strand breaks induced in DNA molecule by radiation of proton and Secondary particles using Geant4 code

Radiotherapy using various beams is one of the methods for treating cancer, Hadrons  used   to  treat cancers  that  are  near critical organs. The most important part of the cell that is damage by ionizing radiation is DNA. In this study, damages induced in the  genetic material of  living cells (DNA) defined by  the  atomic model from the  protein data bank (PDB) have been studied by  radiati...

Induction of Apoptosis on K562 Cell Line and Double Strand Breaks on Colon Cancer Cell Line Expressing High Affinity Receptor for Granulocyte Macrophage-Colony Stimulating factor (GM-CSF)

Background: Immunotoxins are comprised of both the cell targeting and the cell killing moieties. We previously established a new immunotoxin, i.e. Shiga toxin granulocyte macrophage-colony stimulating factor (StxA1-GM-CSF), comprises of catalytic domain of Stx, as a killing moiety and GM-CSF, as a cell targeting moiety. In this study, the ability of the immunotoxin to induce apoptosis and dou...

The Role of Long Non Coding RNAs in the Repair of DNA Double Strand Breaks

DNA double strand breaks (DSBs) are abrasions caused in both strands of the DNA duplex following exposure to both exogenous and endogenous conditions. Such abrasions have deleterious effect in cells leading to genome rearrangements and cell death. A number of repair systems including homologous recombination (HR) and non-homologous end-joining (NHEJ) have been evolved to minimize the fatal effe...
